5.Abstract: Current ferroptosis inducers effectively kill cancer cells by inhibiting GPX4 but also damage immune cells, limiting their therapeutic potential. In this study, the authors screened over 4,200 small molecules and identified N6F11, a compound that selectively induces GPX4 degradation in cancer cells without affecting immune cells. Mechanistically, N6F11 directly binds the E3 ubiquitin ligase TRIM25, activates its ubiquitin ligase activity, and promotes K48-linked ubiquitination and proteasomal degradation of GPX4, thereby inducing ferroptosis. Unlike conventional GPX4 inhibitors, N6F11 spared dendritic cells, CD8⁺ T cells, NK cells, and neutrophils. In pancreatic cancer mouse models, N6F11 triggered immunogenic ferroptosis accompanied by HMGB1 release, dendritic cell activation, and CD8⁺ T-cell–mediated antitumor immunity. Furthermore, combining N6F11 with anti-PD-L1 immunotherapy markedly suppressed tumor growth and prolonged survival in KRAS/TP53-driven pancreatic cancer models. These findings establish TRIM25-mediated GPX4 degradation as a promising strategy to induce tumor-selective ferroptosis while preserving antitumor immunity.


6.한글 초록 요약본: 기존의 GPX4 억제제(RSL3, Erastin 등)는 ferroptosis를 유도하여 암세포를 사멸시키지만, 면역세포까지 손상시켜 항종양 면역을 약화시키는 한계가 있다. 본 연구에서는 4,208개의 화합물을 스크리닝하여 N6F11이라는 새로운 저분자 화합물을 발굴하였다. N6F11은 암세포에서만 GPX4 단백질을 선택적으로 분해하고 면역세포에는 거의 영향을 주지 않았다. 기전적으로 N6F11은 TRIM25 E3 ubiquitin ligase의 RING domain에 직접 결합하여 TRIM25 활성을 증가시키고, GPX4의 K48-linked ubiquitination과 proteasomal degradation을 촉진하여 ferroptosis를 유도하였다. 또한 HMGB1 방출을 증가시켜 dendritic cell과 CD8⁺ T 세포를 활성화하고 강한 항종양 면역반응을 유도하였다. 췌장암 마우스 모델에서는 종양 성장을 효과적으로 억제하였으며, anti-PD-L1 면역항암제와 병용 시 치료 효과와 생존기간이 더욱 향상되었다. 따라서 N6F11은 면역세포를 보존하면서 암세포에서만 ferroptosis를 유도하는 새로운 치료 전략으로 제시되었다.


7.한글 논문 요약본

7-1 Introduction : 면역관문억제제(PD-1/PD-L1)는 암 치료에 큰 발전을 가져왔지만 대부분의 환자에서는 충분한 치료 효과를 얻지 못한다. 효과적인 항종양 면역을 위해서는 암세포의 면역원성 세포사(ICD)가 중요하며, HMGB1과 같은 DAMP의 방출이 dendritic cell을 활성화하여 CD8⁺ T세포 면역반응을 유도한다. Ferroptosis는 GPX4 억제를 통해 발생하는 세포사로 암 치료의 새로운 전략으로 주목받고 있으나, 기존 GPX4 억제제는 암세포뿐 아니라 면역세포에서도 ferroptosis를 유도하여 오히려 항암면역을 저하시킨다. 이에 저자들은 암세포에서만 선택적으로 GPX4를 제거하는 새로운 화합물을 개발하여 항암 효과와 면역반응을 동시에 증강시키고자 하였다.


7-2 Method : 연구진은 4,208개의 합성 화합물을 대상으로 In-Cell Western screening을 수행하여 GPX4를 감소시키는 후보물질을 선별하였고, 그중 N6F11을 선정하였다. 다양한 암세포주와 면역세포에서 GPX4 발현, ferroptosis, 세포생존율을 비교하였으며, ubiquitination assay, immunoprecipitation, CETSA, molecular docking 등을 통해 TRIM25와 GPX4의 상호작용을 분석하였다. 또한 TRIM25 knockdown 및 GPX4 mutant를 제작하여 기전을 검증하였다. 이후 KPC 췌장암 마우스 모델과 KRAS/TP53 유전자변이 모델에서 N6F11 단독 또는 anti-PD-L1과 병용 투여하여 종양 성장, HMGB1 분비, CD8⁺ T세포 활성 및 생존기간을 평가하였다.


7-3 Result : 4,208개의 화합물 스크리닝 결과 N6F11이 가장 강력하게 GPX4 단백질을 감소시키는 후보물질로 확인되었다. N6F11은 GPX4 mRNA에는 영향을 주지 않고 단백질만 선택적으로 분해하였으며, proteasome 억제제 처리 시 이 효과가 사라져 ubiquitin-proteasome system을 통한 분해임이 확인되었다. 또한 TRIM25가 GPX4의 새로운 E3 ligase로 작용하며, N6F11은 TRIM25의 RING domain에 직접 결합하여 GPX4의 K48-linked ubiquitination을 촉진하였다. GPX4 K48R mutant에서는 N6F11에 의한 ferroptosis가 거의 유도되지 않아 해당 기전이 필수적임을 입증하였다.


기능적으로 N6F11은 암세포에서는 강한 ferroptosis를 유도했지만 CD8⁺ T세포, NK세포, dendritic cell, neutrophil 등 면역세포에는 거의 독성을 나타내지 않았다. 췌장암 마우스 모델에서는 HMGB1 방출 증가와 dendritic cell 활성화, CD8⁺ T세포 의존적인 항종양 면역을 유도하여 종양 성장을 효과적으로 억제하였다. 특히 anti-PD-L1 항체와 병용 투여했을 때 단독 치료보다 종양 성장이 더욱 감소하고 KRAS/TP53 변이 췌장암 모델의 생존기간이 약 2.5배 연장되었다. 반면 HMGB1 또는 CD8⁺ T세포를 제거하면 이러한 효과가 소실되어 N6F11의 항암효과는 ferroptosis에 의해 유도된 면역반응에 의존함을 확인하였다. 따라서 연구진은 TRIM25-GPX4 축을 이용한 선택적 ferroptosis 유도가 면역항암제의 효과를 높일 수 있는 새로운 치료 전략임을 제시하였다.
